A director can leave the board by resigning, or be removed by the shareholders. Either way, the change must be filed with the ROC so the board composition on record is accurate — minimum-director requirements must still be met afterward.
A resignation involves the director's resignation letter, the board taking note, and filings: DIR-11 (optionally by the director) and DIR-12 (by the company). A removal by members under Section 169 is a more formal, adversarial process needing special notice, a resolution and an opportunity for the director to be heard.
